Chapter 6, Problem 25P
Summary Introduction

To analyze:

Analyze the term genetic complementation, and answer the following questions.

Relate how the term genetic complementation applies to an experiment in which two lysis-defective bacteriophages are able to co-infect a bacterial cell and produce lysis.

Find out another example of complementation in this book, and describe how the genetic complementation applies to this case.

c. Analyze if the genetic complementation has the same meaning in both cases. If yes, explain how.

Introduction:

The term genetic complementation of two genes suggests that the two mutated genes having a mutation at two different locations produce wild type character, therefore completing each other. Complementation is also called Cis-trans mutation. Genetic complementation is used to determine whether mutation has occurred in the same gene or in two different genes.

Flow cytometry analysis was performed on the blood of an individual known to have been recently exposed to both rhinovirus (RV) and Mycobacterium tuberculosis (M.tb; an intracellular bacterial pathogen). Although an effective T cell response against RV was detected, a T cell response against M.tb was absent. Subsequently, genetic testing was performed, and the individual was found to possess two mutations. Below, indicate the MOST LIKELY mutations. A. T-bet; IL-12 B. CD28: IL-2Ra C. RORyT: IL-17 D. CD3: CD4
