Concept introduction:

The Sanger N-Terminal analysis is a method for sequence analysis of amino acid using 2,4-dinitrofluorobenzene (DNFB). The nucleophilic aromatic substitution reaction occurs when a polypeptide and DNFB react in mildly basic solution. The subsequent hydrolysis gives a mixture of amino acids with the N-terminal amino acid labeled with a 2,4-dinitrophenyl group.
