Chapter 19, Problem 20P
Interpretation Introduction

To write:

Post translational modification via Michael addition.

Introduction:

The tricarboxylic acid (TCA) cycle consist of a series of various chemical reactions in order to release the energy stored by the oxidation of acetyl-CoA (acetyl-Coenzyme A) into carbon dioxide and ATP (adenosine triphosphate). It is also called as the Krebs’s cycle.

The Krebs cycle is called as the citric acid cycle as the metabolic pathway is derived from citric acid, which is used as well as regenerated in a series of reactions in order to complete the cycle. The cycle also requires the acetate and water.

The Michael reaction also known as Michael addition in which there is addition of a nucleophile of a carbanion takes place to the a, ß unsaturated carbonyl compound.

A Leu →Ala mutation at a site buried in the core of the enzyme lysozymeis found to be destabilizing. Explain the observed effect of this mutationon lysozyme stability by predicting how enthalpy (ΔH°), conformationalentropy (ΔS°peptide), and the hydrophobic effect (ΔS°solvent) are expected to change for the mutant compared to wild-type lysozyme. Explain how ΔG°for unfolding is affected by your predicted changes in enthalpy or entropy.
