Since only one out of two samples has plasmid DNA. The sample with DNA of other forms can be single-stranded or double-stranded, readily cleaved by DNase I. Whereas the one with plasmid DNA, the highly supercoiled, is not easily affected by DNase I.
In a few reports, there are instances of plasmid DNA acted by DNase I, but it depends on the ionic strength of the buffer and treatment duration.
