Chapter 6, Problem 18P

An interrupted mating study is carried out on Hfr strains 1, 2, and 3 identified in the problem above. After conjugation is established, a small sample of the mixture is collected every minute for 20 minutes to determine the distance between genes on the chromosome. Results for each of the three Hfr strains are shown below. The total duration of conjugation (in minutes) is given for each transferred gene.

Chapter 6, Problem 18P, An interrupted mating study is carried out on Hfr strains 1,2,and3 identified in the problem above.

a. For each Hfr strain, draw a time-of-entry profile like the one in Figure 6 .8 a.

b. Using the chromosome map you prepared in answer to Problem 15 , determine the distance in minutes between each gene on the map.

c. Explain why azi is the last gene of strain 2 to transfer in the 20 minutes of conjugation time. How many minutes of conjugation time would be needed to allow the next gene on the map to transfer from Hfrstrain 2 ?

d. Write out the interrupted mating results you would expect after 20 minutes of conjugation for Hfr strains 4 and 5 . Use the format shown at the beginning of this problem.

e. In minutes, what is the total length of the chromosome in the donor species?

An Hfr strain that is hisE + and pheA + was conjugated to a strain that is hisE − and pheA −. The conjugation was interrupted at different times, and the percentage of recombinants for each gene was determined by streaking on media that lacked either histidine or phenylalanine. The following results were obtained:   A. Determine the map distance (in minutes) between these twogenes.B. In a previous experiment, it was found that hisE is 4 minutesaway from pabB and that PheA is 17 minutes from pabB. Drawa genetic map showing the locations of all three genes.
An interrupted mating study is carried out on Hfr strains 1, 2 and 3. After conjugation is established, a small sample of the mixture is collected every minute for 20 minutes to determine the distance between the genes on the chromosome. Times of entry for each of the three Hfr strains studied in the interrupted mating are provided in parenthesis. The total duration of conjugation (in minutes) is given for each transferred gene. Four Hfr strains are derived from an F+ strain of E. Coli to serve as donors for an interrupted-mating experiment. Use the time-table and partial map of the F+ strain (shown below) to determine the genes’ respective positions. Keep in mind that the map distances are NOT proportional, only the FIRST 5 markers are indicated per strain, and the entry times, recorded in minutes, are in parentheses. Transferred genes represent wild-type alleles. Based on the data, which gene can be located at position 5 on the map?
