Chapter 7, Problem 2Q
Summary Introduction

To review:

The roles of OmpR and EnvZ.

Introduction:

A single-celled organism like Escherichia coli has to survive in harsh conditions. For this, they activate different genes according to the situation. For example, expression of the high amount of OmpC in hypertonic solutions as compared to isotonic or hypotonic solutions. Other proteins like membrane protein (EnvZ) get phosphorylated by protein kinase in the signal transduction.

The incubation of cytoplasm protein (OmpR) and membrane protein (EnvZ) with 32P (phosphorus)-radiolabeled ATP is already given as

Table A

Incubation of EnvZ with 32P-ATP Incubation of OmpR with 32P-ATP
Incorporation of 32P label into protein High Absent

The incubation of P-EnvZ with OmpR and testing the samples at different time intervals give following results:

Table B

Duration after mixing 32P-EnvZ and OmpR (min)
0 2 5 10
Presence of 32P label in EnvZ High Medium Low Absent
Presence of 32P label in OmpR Absent Low Medium High
