Chapter 35, Problem 7TYK
Summary Introduction
Introduction:
The roots nodules present in the legume plants are characteristically formed as a result of a mutualistic relationship with bacteria involved in nitrogen fixation (Bradyrhizobium and Rhizobium). The relationship is formed as the nod gene products of the bacteria that permits it to infect the cells of root cortex and then proliferate.

In this activity you will analyze the results of experiments that investigate nutritional requirement of
several mutant strains of yeast. The mutations in these strains cause a nutritional requirement for an
amino acid, such that the strains will not grow in media that lack one specific amino acid. Any mutant
that has a nutritional requirement is called an auxotroph, and is incapable of growing in a "minimal
medium" containing only a carbon source (e.g., glucose), a simple nitrogen source (e.g., ammonium
sulfate), and various salts and minerals. Such strains can be supported on a medium supplemented
with only the missing nutrient or on a "rich" medium that contains amino acids, vitamins, nitrogenous
bases, etc. (often in the form of an extract from yeast). The wild-type individual that can synthesize
the metabolic component is a prototroph, and is capable of growth on minimal medium.
The mutant strains in this activity are unable to synthesize tryptophan, lysine, or histidine; one…
Select the correct statement regarding plant transformation methods.
a.
All statements regarding plant transformation are correct
b.
Electroporation involves shocking plant cells with heat so pores open in the plasma membrane to allow DNA to go into the cell
c.
Agrobacterium-mediated transformation involves bacteria inserting a segment of DNA into a plant's nucleus
d.
The gene gun propels bacteria into plant cells that then insert DNA into the plant's nucleus
According to data from genome sequences, cytoplasmic dynein is absent in some plants (e.g.,Arabidopsis) and present in others (e.g.,rice).Does this finding surprise you? What else might you do to confirm or refute such a statement? How is it possible that higher plant cells could operate without cytoplasmic dynein?

During infection, a plasmid called Ti is transferred into the plant cells, where that plasmid DNA is integrated into the plant’s genome. Some of the genes encoded by this plasmid are expressed by host cell machinery in the plant cell. These gene products produce plant hormones which stimulate cell division in the infected cells, causing the development of a plant tumor which provides an environment for that bacterium to grow. You are studying a novel isolate of A. tumefaciens and want to determine which of the genes on the Ti plasmid are expressed in the bacterium, and which are expressed in the plant.
