To compare: The melting temperature of 1-kb segment of DNA containing 20%. A residue to that of 1-kb segment of DNA containing 30%. A residue under same conditions.
Concept introduction: Melting temperature (Tm) is the temperature at which half of the DNA melts or denatures due to the breakage in the hydrogen bond that holds two strands together. Melting temperature of a DNA duplex makes changes in the structure and stability of the DNA. Melting temperature of a DNA double helix depends on the number and percentage of base-pairs present in the structure. More the G-C base-pairs present in the structure, stronger the bond and the higher would be the melting temperature to break the bond.
