Procedure:
1. Label 5 test tubes from 1 to 5. Place the substances as indicated below.
Test tube 1 - 2ml gelatin + 1ml water
Test tube 2 - 2ml gelatin + 1ml (boiled) pineapple juice
Test tube 3 - 2ml gelatin + 1ml pineapple juice
Test tube 4 - 2ml gelatin + 1ml pineapple juice + 5drops 0.25 M MgCl2
Test tube 5 - 2ml gelatin + 5mg Meat tenderizer + 1ml water
a. A purified preparation of papain may be used instead of the meat tenderizer.
b. Test tube 2 should be prepared by putting 1 ml of pineapple juice in the test tube and placing it
in a boiling water bath for 5 minutes. The tubes should then be allowed to cool to 50°C before
proceeding.
2. Mix the contents of the test tubes and place them in a water bath kept at 40°C – 50°C.
3. Keep the tubes in the water bath for 5 minutes, shaking once or twice to promote mixing. Air-cool
for 5 minutes, then place in an ice bath. Observe for evidence of setting or solidifying.
4. Record the results.
