The GAPDH active site cysteine 149 was changed to serine, resulting in a 104 reduction in the enzyme's turnover rate. K_cat is dramatically reduced by changing a catalytic cysteine, whose nucleophilicity is enhanced by the nearby presence of a His residue, into a serine.
