Converting a catalytic cysteine into a serine or vice versa causes a significant decline in enzyme activity in GAPDH. The GAPDH active site cysteine 149 was changed to serine, resulting in a 104 reduction in the enzyme's turnover rate. Kcat is dramatically reduced by changing a catalytic cysteine, whose nucleophilicity is enhanced by the nearby presence of a His residue, into a serine.
