Clinical Laboratory Techniques 1- Lab Urinalysis on an Abdominal Urine and Centrifuge Destinee Mills Introduction A centrifuge is used to separate materials that have different densities by applying force and rotating balanced units at various speeds. The force and speed required to separate materials depends on the size and viscosity of the specimens. A clinical centrifuge tends to operate at slower speeds and relative centrifuge force (RCF) and accepts a range of heads and cups to accommodate the various array of laboratory specimen’s containers such as serum tubes, bacteria bottles, blood bags, and urine cups/urinalysis tubes. Specifically, the separation of urine by centrifugation is performed so that the liquid within the urine can be separated to reveal any solid components that may be in the sample such as crystals, blood, or microorganisms. The purpose of this lab was to learn how to use a universal tabletop centrifuge and prepare two abnormal urine samples from that can be evaluated and compared to one another. Materials Universal Centrifuge Disposable gloves Clinitest Dipstick Refractometer Acetone Transfer pipettes Water Urinometer Urine sample C Urine sample B Acetest Tablets Clinitest Tablets 20% Sulfosalicylic Acid Test tubes Microliter Pipette Absorbent Mats Kim Wipes Dry paper towels Procedures 1. Prepare a clinical centrifuge for the use, centrifuge the urine samples, centrifuge a sample, and separate the specimens. 2. Perform a routine urinalysis (physical and chemical) test on abnormal urines C & B and record all results. 3. Perform Clinitest 4. Perform Acetest 5. Perform SSA test 6. Perform Ictotest 7. Properly use a automated instrument to analyze urine specimens (Clinitek) 8. Properly use a refractometer to check the specific gravity of urine. 9. Record all test results as they obtained from the performed tests.
