Chapter 8, Problem 4CHI

The death of Geobacillus stearothermophilus spores is routinely used to assess the effectiveness of sterilization procedures. In this procedure, endospores are exposed to the sterilization process (e.g., autoclaving) and then plated on germination medium. If sterilization has been successful, no growth occurs. This requires about 2 days Yung and Ponce have developed a new approach that yields results within 15 minutes after sterilization. Their technique is based on the release of dipicolinic acid (DPA) from the spore coat by germinating cells. DPA release is monitored microscopically after endospores are placed in media containing alanine and terbium ion (Tb3+). Alanine triggers germination and Tb3+ binds DPA. which then fluoresces green when illuminated with UV light. Design an experiment in which this new method could be shown to be as reliable as the culture-based approach. What other sterilization procedures besides autoclaving could this technique be used to monitor?

The agent of gas gangrene is classified as an based on its oxygen sensitivity so in addition to antibiotics and debrideent, hyperbaric oxygen is commonly used in the treatment of gas gangrene. This organism can persist in the outside environment under extreme harsh conditions which include the presence of high levels of atmospheric oxygen because it is able to form an (seen as the absence of color in the Gram stain) which will germinate when introduced into environments such as deep muscle tissue with minimal levels of oxygen, lots of water, and lots of nutrients.
Describe the appearance of an S. agalactiae colony grown on blood agar. Describe how that colony would differ in appearance from a colony of S. pyogenes.
1(a) 1. What is the theory upon which intermittent sterilization is based? (B) Is dry or moist heat more efficient as a sterilizing agent? Why? (C) When would it be necessary to use aseptic techniques when inoculating a tube of nutrient broth with dirt? (D)Why do some microorganisms characteristically form a growth of the pellicle type? What environmental factors might alter the formation of a pellicle? (This is not a graded assignment)
