Chapter 8, Problem 10TYU

DRAW IT The following diagram represents an experiment with isolated thylakoids. The thylakoids were first made acidic by soaking them in a solution at pH 4. After the thylakoid space reached pH 4, the thylakoids were transferred to a basic solution at pH 8. The thylakoids then made ATP in the dark. (See Concept 2.51&1 to review pH.)

Draw an enlargement of part of the thylakoid membrane in the beaker with the solution at pH 8. Draw ATP synthase. Label the areas of high H· concentration and low W concentration. Show the direction protons flow through the enzyme, and show the reaction where ATP is synthesized. Would ATP end up in the thylakoid or outside of it? Explain why the thylakoids in the experiment were able to make ATP in the dark.

A key experiment that helped put doubts to rest regarding the chemiosomotic theory used an artificial vesicle, a light activated proton pump, and ATP synthase. Which of the following are true about this experiment? (select all that apply) Group of answer choices The inside of the vesicle is more negatively charged when the light is turned on ATP production would decrease if the proton pump was not included in the experiment Adding UCP1 into the experiment would increase ATP production ATP production increases when the light is turned on The pH inside the vesicle decreases when the light is turned on   The final stage of the electron transport chain is the transfer of the electrons to ATP synthase. Group of answer choices True False   Complex I can accept electrons from NADH using binding sites on both the matrix and intermembrane space sides of the inner mitochondrial membrane Group of answer choices True False   Different iron-sulfur clusters can have different…

In a photosynthetic investigation, a student measured the following pH of the stroma and the inner thylakoid compartments of spinach leaf cells at the conclusion of the experiment. Source of Fluid pH (mean) Standard Error of Mean (SEM) Stroma 8.2 0.75 Inner compartment of thylakoid 5.6 0.35 Explain the reason for the differences in pH between the stroma and the thylakoid lumen.
