Introduction:
Microscopy started with the basic light microscope and with the passage of time, it has developed into various advanced types. The different types of advanced microscopy techniques are associated with the visualization of a range of structures. The resolution and the detection power of each type of microscopic technique may vary in many aspects.
Fluorescence microscopy is used to detect intracellular events that occur inside a bacterial cell. For example, replication with the help of colored dyes that can absorb and emit light of specific wavelengths. Darkfield microscopy is used for distinguishing certain extracellular features of bacteria like flagella by reducing the intensity of incident light.
The specimens or objects that have dimensions less than that of the wavelength of light can be visualized by both fluorescence and dark-field microscopy. Both of these techniques are able to detect such objects/specimens but these techniques are not able to resolve them. The differences between fluorescence and dark-field microscopy are mentioned below:
