Chapter 20, Problem 24EQ
Summary Introduction

To analyze:

The presence of repetitive sequences, such as transposable elements, that are present in multiple copies within the chromosomal DNA of an organism can be detected by Southern blotting. In Southern blot, in a given figure, chromosomal DNA was isolated from three different strains of baker’s yeast, digested with a restriction enzyme, run on a gel, blotted, and then labeled with a probe that is complementary to a transposable element called the Ty element.

Genetics: Analysis and Principles, Chapter 20, Problem 24EQ

Using the above information, explain why the banding patterns are not the same in lanes 1,2,3.

Introduction:

Southern blotting is a blotting method that uses a labeled DNA probe to detect the presence of a particular gene sequence within a mixture of many gene sequences. Transposable elements are a family of genes that include sequences of DNA that moves from one location in the genome to another. Yeast transposable element (Ty) is a family of genes that contains repetitive DNA sequences. These transposable elements can be detected by Southern blotting technique.
