Evaluation at Atomic Resolution of the Role of Strain in Destabilizing the Temperature Sensitive T4 Lysozyme Mutant Arg96-->His
T4 lysozymes helps to release the phage particles synthesized inside the host cell via hydrolyzing the 1-4 beta glycosidic bond between N-acetyl D glucosamine and N-acetyl muramic acid making up the peptidoglycan layer of host cell wall. T4 lysozymes are one of the most studied proteins to get a better understanding of folding, stability and activity of proteins.
Point mutations to the T4 lysozyme could stabilize or destabilize the protein. Point mutations that end up stabilizing the protein even more, tend to increase the proteins melting point (Tm) to 3-4 degrees ( 5.1 degrees maximum ) whereas destabilizing point mutations can reduce the Tm by up to 20 degrees.
Point mutations in the 96 the residue of T4 lysozymes are generally destabilizing in nature.
