Question

. In many bacterial species, regulatory sRNAs have
been identified by transcriptome sequencing (RNASeq). How do researchers know that the small RNA
species identified by cDNA sequencing are regulatory
sRNAs rather than fragments of longer mRNAs?

Expert Solution
Step 1Analysis

Once the whole RNA is extracted and cDNA is synthesized, the cDNA is then sent for sequencing. Once the sequenced data comes the sequenced data is being compared with the available data bases of sRNA which can be done with different bioinformatic tool. Those short RNAs which are matching with the existing data are sRNA while others are just parts of long mRNAs.

Step 2 List of Databases
  • BSRD is a repository for published sRNA sequences with multiple annotations and expression profiles.
  • SRD  is a database of Staphylococcus aureus sRNAs with sequences, predicted structures, and genome start and end sites.
  • sRNAdb  is a databse of sRNAs from Gram-positive bacterial species with sequence annotation.
