MacConkey agar acts as a differential medium for lactose fermenting bacterium and non-lactose fermenting bacterium. If the agar retains the red colour and the colonies are opaque, then they are lactose fermenters and if the agar is colourless and the colonies are translucent, then they are non-lactose fermenters.
