Chapter 26, Problem 1S
E. coli lysogens derived from infection by phage λ can be induced to form progeny viruses by exposure to radiation. The inductive event is the destruction of a repressor protein that keeps the prophage genome unexpressed. What might be the normal role of the protein that recognizes and destroys the λ repressor?

M13 is a filamentous phage that infects the bacterium Escherichia coli. Infection with M13 is
not lethal. However, the infection causes turbid plaques in E. coli because infected bacteria
grow slower than the surrounding uninfected bacteria. This phage has been engineered to act
as a vector system. Explain how the amplification of gene of interest works in this phage with
illustration.
Consider the phase variation of flagella in Salmonella. Describe the phenotype of a cell in which hin is deleted. Will it be motile? Will it undergo phase variation? Which flagellin(s) will be used, if any? Name two bacterial defenses against bacteriophages and explain how they work How might a phage evolve resistance to the CRISPR host defense?
The linear dsDNA genome of λ binds on the LamB receptor of E. Coli and conducts a normal lysogenic cycle. Exposure to stress will cause the excision of λ prophage from the E. Coli genome. The excised λ genome is then replicated, packaged, and released from the cell as mature λ phage particles and ready to infect other bacterial cells. Among λ phage particles, the transducing phage mediates a specific type of recombination. Understand this scenario and answer the following questions.
1. What are the basic requirements for the insertion of λ into the E. Coli genome? 2. What special features are found in the λ insertion site? 3. What type of recombination occurs with λ insertion in the E. Coli genome? 4. How you will differentiate λ transducing phage from normal λ phage? 5. What exclusive mechanism λ phage utilizes for recombination?

- The size of a DNA fragment that can be inserted into an unmodified λ vector is very limited. Large segment in the central region of the λ DNA molecule can be removed without affecting the ability of the phage to infect Escherichia coli cells. However, the deletion of this non- essential region does affect the life cycle of the bacteriophage. Explain this statement.arrow_forwardWhen various strains of λ phage are seeded on a lawn of E. coli, they can form clear or turbid plaques. - What is the difference between the lytic and lysogenic cycle of bacteriophages? In the lysogenic cycle, the phage DNA integrates itself into the host genome after penetration, and continues to biosynthesis when conditions change The host cell is not killed in the lysogenic cycle The lytic cycle occurs in the nucleus, while the lysogenic cycle occurs in the cytoplasm In the lysogenic cycle, penetration is achieved through phagocytosisarrow_forwardIn Hershey-Chase experiment, bacteriophages protein coats were tagged with radioactive isotope S-32. - Which of the following is a known characteristic or procesds that can lead to genetic changes in some viruses? When two different viral strains of the same type of virus infect a single individual, reassortment/mixing of segmented genome pieces can occur, resulting in new combinations of segments. Genetic rearrangements of the viral genome occurs as a result of duplicate operons in the viral genome undergoing homologous recombination. After this, if the bacterial cell that this lambda phage infects is robustly healthy, it will make many proteases that overwhelm the efforts of the clll protein in protecting the cll protein from degradation. If the cll protein is degraded, protein will block transcription from the PRM promoter and promote the lysis the cro pathway.
