The linking of fragment of DNA can be done with the help of an enzyme DNA ligase. Each nucleotide get attached as substrate to DNA during the replication for which DNA ligase comes into play.

DNA ligase is a class of enzyme used to ligate or add fragments of DNA for the purpose of repairing or replication. It joins the nucleotide by creating phospho diester linkage.
