Chapter 23, Problem 1P
Interpretation Introduction

Interpretation:

The reason why denaturation of proteins makes them better substrates for proteolysis should be determined.

Concept Introduction:

Proteolysis can be defined as the breakdown of peptides or proteins into amino acids by the action of enzymes. It is classically catalyzed by the proteases that are cellular enzymes. It occurs by intra-molecular digestion. Denaturation of proteins means the degradation of proteins to a simpler level.

Answer to Problem 1P

When the proteins become denatured, they are more accessible by the proteolytic enzymes as substrates for the proteolysis process than are natural proteins.

Explanation of Solution

A protein has a 3-dimensional structure that is not accessible to the proteolytic enzymes. It happens because the proteolytic enzymes cannot act on the peptide bond when the proteins become denatured. Denatured proteins are therefore more accessible as substrates for the proteolysis process than are the natural proteins.

Please help
You have isolated a protein and determined that the native molecular weight of the holoenzyme is 160 kD using size exclusion chromatography. Analysis of this protein using SDS-PAGE revealed 2 bands, one at 100 kD and one at 30 kD. Describe the architecture of the polypeptide component of this enzyme.
