Chapter 19, Problem 19P
The following figure shows the temperature-shift analysis of C. elegans embryos from mothers homozygous for a temperature-sensitive allele of the zyg-9 gene, which helps determine the polarity of the early embryo. Each dot represents an individual embryo subjected to a short (5-minute) pulse of high temperature. Green dots indicate that the embryo ultimately survives and becomes a normal worm, whereas red dots represent animals with abnormal polarity that ultimately die.
a. | At what time after fertilization is the zyg-9 gene required for normal development of C. elegans? |
b. | The same results were obtained whether the sperm used in the fertilization had wild-type or mutant alleles of zyg-9. What does this fact say about the source of the Zyg-9 protein that is present during the narrow window of time when it is required? |

Figure 13-7 illustrates the expression of the Ultrabithorax(Ubx) Hox protein in developing flight appendages. What is the relationship between where the protein isexpressed and the phenotype resulting from the loss ofits expression (shown in Figure 13-1)?
Here are schematic diagrams of mutant Drosophila larvae.
The left side of each pair shows a wild-type larva, with gray boxes showing the sections that are missing in the mutant larva. Which type of gene is defective in each larva: a gap gene, a pair-rule gene, or a segment-polarity gene?
A C. elegans (nematode) gene called par-1 helps todetermine the AP axis of the animal early in development. Scientists determined that par-1 is pleiotropic—it also has a later function in forming the vulva of theadult animal. How could researchers circumvent thelethality of par-1− mutants to observe the later function of the par-1 gene? (Hint: C. elegans larvae caneat bacteria expressing RNAi for any gene.)

- Suppose a researcher has three different Drosophila strains that have mutations in the bicoid gene called bicoid-A, bicoid-B, and bicoid-C; the wild type is designated bicoid +. To study these mutations, phenotypically normal female flies that are homozygous for the given bicoid mutation were obtained, and their oocytes were analyzed using a Northern blot to determine the size and/or amount of the bicoid mRNA and in situ hybridization to determine the bicoid mRNA location within the oocyte. A wild-type strain was also analyzed as a control. In both cases, the probe was complementary to the bicoid mRNA and the results are shown below. (Anterior is on the left; posterior is on the right.)
