Hershey and Chase performed the experiment to determine the transforming material of bacteriophage by using radioactive 32P and 35S. Presence of 32P in the pallet confirmed the presence of DNA in the phage as phosphorous is the constituent of DNA.

Introduction:
Many scientists tried to identify the genetic material of the organisms, but most of them failed to conclude. The experiment conducted by Hershey and Chase is widely accepted and the result obtained satisfied all the criteria of the genetic material. They used the unique constituents of DNA and protein to perform their experiment. P is unique in DNA and S is unique in proteins, so they used radioisotopes of P and S to easily identify the presence in the progeny.
