![5) One very common way to do a preliminary test for whether or not a compound (a new drug, a sunscreen, an artificial
sweetner, whatever) causes mutations, before doing toxicity testing in cultured cells or mice, involves using E. coli that
have lost the ability to synthesize the amino acid histidine for themselves. These his E. coli grow just fine as long as the
growth medium they are cultured on contains histidine, but if they are cultured on medium that does not contain
histidine, they will rapidly die due to lack of that essential amino acid. This is called the Ames test, after the scientist,
Bruce Ames, who developed it.
To test the mutagenic properties of IP-1383, a novel derivative of ibuprofen being analyzed as a potential anti-
inflamatory drug for the treatment of rheumatoid arthritis, his E. coli were incubated in an aqueous solution of IP-1383
for 4 hours and then plated on growth medium that did not contain histidine. As a control, genetically identical his¨ E.
coli were incubated in sterile water and plated on the same growth medium. To test whether or not IP-1383 is toxic to E.
coli cells, another sample of the his E. coli that were incubated in IP-1383 were plated on growth medium that
contained histidine. After the E. coli were allowed to grow and form colonies on the growth medium for 18 hours, the
number of colonies on each plate was counted and the following results were obtained:
