p-Toluidides and Anilides: Heat a mixture of the acid (200 mg) and thionyl chloride
(1 mL) in a medium reaction tube for 30 min in a 50°C water bath. Cool the reaction
mixture in an ice bath, and add 0.5 g of aniline or p-toluidine in 4 mL of toluene
dropwise. Warm the mixture on the water bath for 2 min and wash with 1-mL portions
of water, 5% hydrochloric acid, 5% sodium hydroxide, and water. Pipette out the organic
(top) layer into a clean test tube after each washing step. The toluene is dried briefly
over anhydrous calcium chloride pellets and evaporated in the hood; the derivative is
recrystallized from water or ethanol-water.
