The SDS-Polyacrylamide Gel Electrophoresis (SDS-PAGE) of a protein yields two bands corresponding to molecular weights of 15,000 Da and 35,000 Da. In a separate experiment, the total molecular weight of the native protein was determined to be 85,000 Da. Determine how many of each subunit are present in the native protein. [3 points] There would be two 35,000 Da and one 15,000 Da because 35,000 + 35,000 + 15,000 = 85,000 Da. 4. Explain why beta-mercaptoethanol (BME) is used in denaturing proteins. [2 points] BME is used to reduce the disulfide bonds so that it can irreversibly denature proteins. 5. Explain why SDS-PAGE gel needs to be rinsed in ultrapure water three times for five minutes each before the gel is stained in Coomassie Blue dye solution. [3 points]
