Interpretation:
In the experiments of Barrick et al. it was observed that replacement of histidine by a noncovalently bonded imidazole not only reduced cooperativity but also increased the oxygen affinity of the hemoglobin.
Concept Introduction:
Hemoglobins have two α chains and two β chains. When O2 binds hemoglobin, there is a transition in the hemoglobin structure from the low-affinity, tense (T) state to the high-affinity, relaxed (R) state. Binding of oxygen to hemoglobin follows an allosteric mechanism.
