Concept explainers
(a)
To determine: The concentration of NaCl in protein sample once dialysis has come to equilibrium assuming that no volume change occurs in the sample during dialysis.
Introduction:
Dialysis is the process used to separate the protein molecules from small solutes. The separation takes place because of the larger size of the protein. The partly purified protein extract is placed in a tube composed of a semi permeable membrane. Dialysis mainly retains the larger protein molecules within the membranous tube. It allows the other solute concentrations in protein separation to change until they come into equilibrium with the solution at the outer membrane.
(b)
To determine: The final NaCl concentration in the sample if original 1mL sample were dialyzed twice successively against 100mLof the same Hepes buffer with 0mM NaCl.
