Summary Introduction

The recombinant proteins are purified by a common method called polyhistidine tagging or His-tagging. This process involves the addition of a series of histidine residues either to the C- or N- terminus of the protein. The amino acid Histidine is used for tagging because it has a high affinity for the metal ions. Proteins having multiple residues of histidine will preferentially bind to a solid phase material called resin which has exposed to cobalt or nickel atom. This allows the separation of His-tagged proteins from the other cellular constituents.
