Primase is a RNA polymerase that synthesizes short RNA sequences called “primers”. Primers are complementary strands of a single stranded DNA (ssDNA) template. Primers incorporate dNTPs on the template on the 3 OH group that can attach to the incoming dNTPs. Primers are required by DNA polymerase to add DNA nucleotides.

Introduction:

The rolling circle method of DNA replicationdemonstrates a unidirectional replication of nucleic acid which synthesizes multiple copies of DNA or RNA rapidly. For example, plasmids, bacteriophages, and the genome of virioids. In this replication,“the outer strand works as a positive strand and the inner strand works as negative strand”. DNA replication begins to start by creating a nick (cut) at positive strand. The strand has served as a template for the strand. The negative strand serves as a template for making the  strand.
