To explain: The portion of the nuceloplasmin molecule responsible for localization in the nucleus.
Concept introduction: Nuclear membrane has nuclear pores bounded by the nuclear pore complexes or the nucleoporins. The nucleoporins allow bi directional movement of the proteins, ions between the nucleoplasm and the cytoplasm. Nucleoplasmin is a nuclear protein nucleoporin which is a molecular chaperone and also involved in chromatin assembly. The nucleoplasmin is translated in the cytoplasm. It has nuclear localization sequence ( NLS) which is detected by the nucleoporins and the exported into the nucleus.
To explain: The way in which the experiment distinguishes between active transport in which nuclear localization triggers transport and passive diffusion in which a binding site for a nuclear component allows accumulation in the nucleus.
