Step 1 Introduction

Intracellular target enzymes are triggered by cell surface receptors. Such enzymes are coupled with G proteins and lead to  further downstream signaling. They amplify the signal commenced after ligand binding. In many cases there are a chain of such reactions that are transmitted downstream and regulate various cell processes; a mechanism known as intracellular signal transduction.

Transmembrane proteins or G protein–coupled receptors (GPCRs) couple with G proteins. The signal transduction is initiated with an inactive G protein, which upon ligand binding GPCR undergoes a conformational change to an active G protein. Upon activation, the Gα binds to a GTP, dissociates itself and leads a cascade of signals downstream.

The overall power of the signal amplification caused by a GPCR is driven by the shelf life of both the ligand-receptor complex as well as the receptor-effector protein complex and the deactivation time of the activated effectors and receptors with the help of intrinsic enzymatic activity. For example, via protein kinase phosphorylation or internalization that is b-arrestin-dependent.
