Question

Give examples of two histamine antagonists that have very different tissue specificities and functions.

Expert Solution
Step 1

Histamine is involved in local immune responses(allergies) to a particular substance such as pollens or other allergens. It can act as a neurotransmitter and has a profound role in inflammation.

Step 2

Histamine plays a vital role in localized immunity, such as reaction to specific allergens. Histamine helps our body to get rid of any allergens. Histamine increases the vasodilatation, increases the vascular permeability in the transient phase of the acute inflammatory reaction. This histamine also acts as a chemical mediator in acute inflammation. Histamine stimulates gastric gland secretion, causing an increased secretion of gastric juice of high acidity.

Some of the common examples of histamine antagonists are diphenhydramine (e.g., Benadryl), chlorpheniramine (e.g., Chlor-Tripolon), and loratadine (e.g., Claritin).

Histamine antagonists with tissue specificity are-

  • H1-antihistamine (e.g., cetirizine). It is used to block the activity of the H1 receptors in the mast cells. They can cross the blood-brain barrier.
  • H2-antihistamines (e.g., Ranitidine). It is used to block H2 receptors present in gastric mucosa to block gastric acid secretion.
