Step 1

An ion-exchange column is a chromatography method to separate molecules based on charge. An anion exchange column consists of a matrix that is coated with positively charged groups. Example: Quaternary ammonium salts. The protein with the least electronegative charge will elute out first than one with a higher negative charge. The ionic interaction is stronger for the latter.

The isoelectric point (pI) of the protein determines the charge of the protein. Essentially at the isoelectric point, the charge on the protein is nil or zero. The pI is in turn dependent on the pH of the elution buffer pH and the amino acid composition of the protein. The protein is negatively charged at a pH greater than its pI and vice-versa.
