Question
**PCR Amplification of a Small DNA Target**

Here is a particular small target that researchers wanted to amplify using PCR - it’s only about 190 base pairs in length.

Normally, primers would be about 20 nucleotides (nt) in length, but let’s make them short to make this question easier to work with.

Below is the sequence that the researchers entered into their computer (one strand only):

5’-CTTTA-------------------180 nt-------------------GAATT-3’

**Which primer pair would work in their PCR reaction?** *(Note that the primer selection program gave them both of their primers in 5’ -> 3’ orientation to make them easy to purchase.)*

- ⦿ 5’-CTTTA-3’ and 5’-TTAAG-3’
- ⦿ 5’-CTTTA-3’ and 5’-AATTC-3’
- ⦿ 5’-GAAAT-3’ and 5’-TTAAG-3’
- ⦿ 5’-GAAAT-3’ and 5’-AATTC-3’

Expert Solution
Step 1

Polymerase chain reaction is the method which is used to amplify small amount of DNA/RNA sample.

The various components required for PCR include a DNA sample, DNA primers, free nucleotides called ddNTPs, and DNA polymerase.

DNA polymerases, which require a template and a primer and synthesize DNA in the 5' to 3' direction,because DNA pol requires the 3'-OH of the existing strand for adding free nucleotides.
