Protocol
1. Close air compressor pressure release valve (turn it parallel to floor)
–
we have a different
compressor now so the picture isn’t correct, but it has the same functionality.
2. Turn on air compressor
3. Power on rheometer (switch on left side)
4. Log into computer
5. Start Rheoplus software (Start Menu->Anton Paar->Rheoplus->Start Rheoplus)
6. In Rheoplus software: File->New->Gel tab->Frequency Sweep:Standard
7. Initialize the rheometer by pressing initialize button if using for first time today
8. Unscrew the white protecting sheath from the rheometer
9. Attach the PP08 probe

being exceptionally careful not to drop or damage it the probe
• Probe is stored in cylindrical padded container
• Align the lines on the probe and the line on the bottom of the connector on the
rheometer (lines shown in picture below)
• Slide up quick disconnect, slide in probe, and slide down quick disconnect
10. Click “Setup Rheometer” button shown below to open the software control panel
The control panel is a new pop-up window

the following should be done on the control panel:
11. Click on Zero Gap button on control panel

rheometer will find the zero distance
12. Use buttons on software control panel to raise rheometer probe enough to insert gel
• You will never use the hardware up and down buttons on the front of the rheometer
13. Place gel on platform and lower probe until contact. Center the gel with the rheometer probe.
14. The gel should be in contact with probe without smashing it. Make sure probe is applying some
normal force. Less than 0.1 N of pressure is needed.
15. The Position field now reads the thickness of your hydrogel.
• Copy that value into the “Meas. position” field on the control panel
• You should also record this in your lab notebook
17. Click on Enter LVE-Strain

it will open a new popup window.
• Enter the strain amount in the “Initial” field: 1% strain is appropriate for most hydrogels
• Set the frequency sweep range to 500 rad/sec to 1 rad/sec so that you can see both
linear and non-linear part of storage and loss modulus curve
• Close the LVE
-Strain window by pressing the OK button
18. Click on Start Test and save the data into a folder on the computer
• Just continue when a warning about the motor comes up
• Test will start with high frequencies and finish at lowest frequencies
• If you watch closely, you can see the probe move back and forth at middle frequencies
i. 1% strain is not very far, but you can see the alignment mark that showed you
how to install the probe into the rheometer move back and forth
19. When test is complete, copy results from results “spreadsheet” at bottom of Rheoplus window
to a Google spreadsheet and save for analysis
20. Use “Setup Rheometer” to open the software control panel. Raise the probe tip slightly and
change samples and repeat 13-16 and 18-19 for each sample
When finished
1. Raise probe to top-most position using the control panel (click Setup Rheometer to open control
panel)
2. Clean probe tip and stand with ethanol and KimWipes
3. Remove probe and carefully return in to padded storage container
4. Screw white protective shroud onto the rheometer
5. Turn off rheometer

a. Open compressor release valve to allow air and water to drain out (this will be loud)
b. Use paper towels to clean up water from air compressor
c. Leave compressor release valve open
