Lab #4 Testing for Macromolecules OBSERVATIONS Sample A. Iodine Solution B. Sudan IV Solution C. Biuret Reagent D. Benedict’s Solution + Heat 1. Protein Solution Light yellow Light red Light purple Light blue 2. Vegetable Oil Light yellow Dark red Very pale blue/clear Light blue 3. Glucose Solution Light yellow Light red Very pale blue/clear Orange 4. Sucrose Solution Light yellow Light red Very pale blue/clear Light blue 5. Starch Grey Light red Very pale blue/clear Light blue 6. Distilled Water Light yellow Light red Very pale blue/clear Light blue QUESTIONS 1. Describe a positive test for protein. Explain how you know. Biuret reagent tests for protein. If something tests positive for protein, the biuret reagent will turn purple. I know this because from the observation table above, all the other solutions when mixed with a protein solution turned light blue, almost clear, but the biuret reagent was the only solution that brought forth a significant color change. 2. Describe a positive test for lipid. Explain how you know. Sudan IV tests for lipid. It was the only one out of the rest of the solutions that completely took in the red colour, unlike the others that were only slightly red. 3. Describe a positive test for glucose. Explain how you know. Benedict’s solution + heat tests for glucose. In a positive test for glucose, benedict’s solution (after being heated) turned orange, the outlying colour as the other solutions all turned light blue. 4. Describe a positive test for starch. Explain how you know. The iodine solution tests for starch. If starch is present, the iodine solution will turn grey, while for the rest of the solutions, their colours were all light yellow. Iodine was the outlier.
5. What was the purpose of testing distilled water for each part of the investigation? The purpose of testing distilled water for each part of the investigation was to see what a negative test result would look like as water has no protein, fat, carbs or starch.
