Fall 2023 Tutorial 1 - Data Analysis & Identification of Mystery Bacteria BIOL 1103 Tutorial 1 Assignment Full Name: Ali Khelil Student #: 101258560 Lab section: A14 Although the tutorial work may be completed in groups, all work submitted must be completed individually. It is important that you do not copy from the lab manual or from each other. DO NOT plagiarize! Unless otherwise specified in the question, all answers should be written using complete sentences, paragraph style. Part I: Analyzing & Interpreting Section Data 1. Manipulating section data in Excel. a. Use the Data Analysis ToolPak to run a One-way ANOVA. You will then perform Tukey’s Test to determine which tre atment pairs are significantly different (consult Appendix 2) . What is your ANOVA p-value and what were your Tukey’s q scores for each treatment (2 marks) ? ANOVA p-value: 2.20855E-07 Raw v. Filtered q score: 6.06401684 Filtered v. Boiled q score: 3.471584475 Raw v. Boiled q score: 9.535601315 b. Plot your data as a boxplot and carefully paste it into space below. Be sure to include properly formatted titles, legend, and axes (3 marks) . Consult Appendix 2 for guidelines on creating graphs.
Fall 2023 Tutorial 1 - Data Analysis & Identification of Mystery Bacteria BIOL 1103 Table 1.0: Box plot of the colony sample size form the Raw, Filtered, and boiled water. c. Were your results significant if your alpha was set to 0.05? What does this mean for your original hypothesis and predictions (2 marks) ? The results were significant since the p-value was less than the alpha. If the p-value was greater than the alpha, then it would mean my hypothesis and predictions were incorrect. d. Provide a brief description of your results as if they were being incorporated into a formal report (2 marks) . Hint : Refer to Appendix 1. The p-value discovered through calculations is 2.20855E-07. The p-value is less than the alpha value of 0.05. The q scores for each colony is 6.06401684, 3.471584475, and 9.535601315 respectively and is shown in depth in table 1.0. 2. Provide another means, and a brief description of the method (max 3 sentences), you could use to sterilize the raw river water (2 marks) . ( Do not forget to include citations and a references section if using outside sources) . Chlorinating the water is another way of sterilizing the raw river water. Chlorine is a powerful chemical that kills bacteria, germs and any other organisms that would float around in a river and would be perfect in a small sample size like a bucket full of water.
Fall 2023 Tutorial 1 - Data Analysis & Identification of Mystery Bacteria BIOL 1103 3. Imagine you are writing the final paragraphs of a discussion section of a lab report. Briefly discuss 2 next steps that you could take to further this study (i.e., enumeration of contaminants, survey of organisms present, varied treatments, etc.) (2 marks) . Hint : Refer to Appendix 1. The next 2 steps that would be done is to comment on any anomalous data that you’ve collected, and you should demonstrate that you understand the meaning of the experiment and the results that were achieved. Part 2: Identification of Mystery Bacteria An individual checks themselves into a local hospital after experiencing severe symptoms of dehydration, diarrhea, and muscle cramping. They recently returned from a weeklong camping trip deep in the Algonquin Provincial Park. They had hiked in with a group of friends and portaged from campsite to campsite throughout their trip. The group brought their own provisions but did not bring any potable water. Instead, they relied solely on the river water next to their campsites. A few days after returning from the trip, the aforementioned symptoms began to present. Once admitted to the hospital, the nurses and doctors ran a series of tests on the individual to determine the source of the illness. They determined that the symptoms were likely the result of a bacterial infection. The doctors managed to isolate the likely bacteria from a stool sample. The bacteria were plated on 2 different agars (a blood agar and a Lysongeny broth agar) and left to incubate for 24 hours. A small colony sample was taken from the Lysongeny broth agar and viewed under an SEM and a compound light microscope at 100X (Figures 5 and 6, respectively). The bacterial agar plating results (Figures 5 and 6) will be provided and examined during your scheduled tutorial session. Your job is to use the above information, in addition to the figures provided during your tutorial session, to determine the identity of the bacteria likely causing the infection. 4. From where did the campers likely contact the bacterial infection (1 mark) ? When they drank the raw river water without cleaning it.

Fall 2023 Tutorial 1 - Data Analysis & Identification of Mystery Bacteria BIOL 1103 5. The image taken at 100X was stained with crystal violet. What is the likely gram type of the species? Provide a brief justification for your identification (2 marks) ? It would be gram positive because the cells would absorb the dye to make them stand out against the background. 6. What is the main method of motility for this species (1 mark) ? The main method of motility is cilia. 7. Using the table provided below, which of the following species is most likely to have infected the individual (1 mark) ? Bacteria Name Size (um) Method of Motility Blood Agar Colony Colour Lysongeny Broth Agar Colony Colour Ehrlicians feber 2 Single Flagellum White Yellow Serratiatus hosta 2-5 Triple Flagellum White Pink Lactococcus snorkraka 2-2- Single Flagellum White Pink Vermontus nysa 1-3 Single Flagellum Yellow Yellow Streptobacillus frossa 1-3 Cilium White Black Streptobacillus frossa would be the likely cause. The following questions may require you to do some research. If you are using an outside source, be sure to add references at the end of your assignment. Refer to Appendix 1 for proper referencing guidelines.
Fall 2023 Tutorial 1 - Data Analysis & Identification of Mystery Bacteria BIOL 1103 8. Would Penicillin or Amoxicillin be better suited to treat the individual’s infection? What is the main method of action used by the antibiotic you would prescribe, and provide a brief justification for your choice (3 marks) ? Both penicillin and amoxicillin are used to treat many of the same bacteria infections but amoxicillin covers a greater range of bacteria so it would be better to use. 9. Besides using antibiotics, what other treatment methods should be prescribed to the individual (1 mark) . Garlic has been researched and tested to treat bacteria infections including salmonella and E. coli. 10. What recommendations would you make for them to prevent future infections (1 mark) ? Clean the water by boiling the water and filtering the water with a fine mesh. References: Leyla Bayan, Peir Hossain Koulivand, Ali Gorji. (2014). (Garlic: a review of potential therapeutic effects). This assignment is due 1 week after completing your Tutorial 1 session, but must be submitted to Brightspace on the hour (i.e., 8 AM or 1 PM, depending on your section) prior to your next session. IMPORTANT : Lab 2 begins the week of Oct. 9 (Group 1) and Oct. 16 (Group 2). Refer to schedule in Brightspace, or ask the lab coordinators if you have any questions.
