Purpose (/2) To determine the INSR inserts orientation using an agarose gel in addition to performing a digest on plasmid pAMP with enzymes EcoR1 and BamHI. Results (/5) Gel results for lane 1 are approximately a size of 11 KB in reference to the top band. For the middle band and bottom band, they are approximately 10 KB and 8 KB, respectively. For lane 2, the band is a size of approximately 6 KB, as well as lane 3, where the band as well is a size of 6 KB. The top band is 5 KB and the bottom band is about 1 KB.

Discussion (/2) When analyzing the electrophoresis gel, orientation B of the double digest reflects a 3’-5’ direction where the INSR is being ligated into the plasmid. Lane 1 yielded 3 bands: one nicked (top), one linear (middle), and one supercoiled (bottom). The supercoiled is the smallest band, meaning it will migrate the easiest through the gel. Both lanes 2 and 3 were analyzed and evidently, both formed a single band. The plasmid size/insert were concluded to be 6 KB of single digests.
