Summary Introduction

To determine: The use of DNA primers complementary to ${\text{5}}^{\prime }$ ending strand (which is identical to ${\text{3}}^{\prime }$-ending strand sequence) which are used to close the gaps at the end of each contig.

Introduction:

A desired DNA segment can be amplified if at least the sequences of its end portion are known by polymerase chain reaction (PCR). PCR uses DNA polymerase enzyme which uses deoxyribonucleotides and synthesize strands of DNA using DNA template. DNA polymerase adds nucleotides to already existing strands called primers. PCR uses primers which targets sequences present at the ends such that their ${\text{3}}^{\prime }$ end is positioned toward the segment which is supposed to be amplified. The PCR primers are complementary to the opposite strand sequences of target DNA.
