To write:
The essential hypothesis made in the method in which the number of bacteria is calculated from the number of colonies.
Introduction:
A culture plate:
In biology, it is a low and flat-bottomed laboratory container. It is useful to grow a layer of organisms on a thin nutrient medium layer. These includes the molds, bacterias, and other cells. The most ordinary types of culture plates are the multiwell plate and the Petri dish.

Explanation of Solution
A colony can be defined as a visible mass of microbes that all originate from a single mother cell. Thus, a colony is formed of a clone of bacteria which are all genetically similar.
The hypothesis:
Bacteria grow rapidly in a pure culture. The cell numbers enhance considerably in a short phase of time. The colony is formed by one or more bacteria. The colony is developed from the live bacteria. These bacteria give rise to the colony when there are favorable conditions and adequate amount of nutrients.
So, a colony is produced by one or more bacteria.
One or more bacteria have formed the colony. The colony is developed from the live bacteria.
