Concept Introduction:

RNA is made of ribose sugar and nucleotides. Periodic acid is an oxidizing agent. It oxidizes the vicinal diols to vicinal dial by breaking the carbon-carbon bond between two alcohol groups. The nucleotide at 3' end contains vicinal diol so the reaction takes place at 3' end of RNA molecule.
