Chapter 20, Problem 16TYPSS
Summary Introduction

To review:

The reason behind the nonworking of two primers which have been purchased for the Polymerase chain reactions.

The way by which one can salvage the experiment of PCR (Polymerase chain reaction).

Introduction:

Polymerase chain reaction (PCR) is an advanced technique by which millions of copies of the desired segment of the DNA (Deoxyribose nucleic acid) can be made with the help of the thermocycler. The PCR consisted of three steps: the denaturation, the annealing, and the extension. In the denaturation step, the unwinding of the double-stranded DNA takes place. During the annealing process, the short strand of bases which is called primer gets attached to the single-stranded DNA. In the extension step, the extension of the primer takes place. A thermostable DNA polymerase enzyme is required for carrying out the reactions. Please refer the table given in the question for sequences of both the primers and the template strand.
