Question
Chapter 17, Problem 2DA
Summary Introduction
To determine: The number of spots that would be observed when human cells in the G2 phase of the cell cycle are subjected to the FISH analysis using the probe recognizing telomeric DNA.
Introduction: Fluorescent in situ hybridization (FISH) is used to identify gross chromosomal rearrangements. It is a molecular cytogenic technique in which fluorescent probes are used to detect DNA sequences.

Method:
Observations of onion root tip squash.
Scan the microscope under the 10x objective. Look for the region that has large
nuclei relative to the size of the cell; among these cells will be found cells displaying
stages of mitosis. Switch to the 40X objective to make closer observations. Since
prophase and prometaphase are difficult to distinguish, classify all these cells as
prophase. Record your observations in the table provided.
Examples are shown in the figure below.

- How long does it take for a cell to take one complete cycle? The answer varies on the type of cell and the species. For instance, a liver cell completes its cell cycle for over 22 hours (Becker and Lane 1965) while other somatic cell lasts to about 18 hours.
