Introduction:

Promoter bashing is a genetic technique used to identify the genetic sequences, which play an important role in the regulation of transcription of a gene. This method uses a clone which contains a coding region of a gene and a region upstream from the core promoter, which has regulatory elements like silencers and enhancers. The effect of promoter bashing on transcription can be easily determined by swapping the coding gene with lacZ (lac operon) gene, which codes for beta-galactosidase whose activity can be measured using enzyme assay.
