The genes of interest are inserted next to the promoter in the plasmid. The open reading frame (shortly ORF) is a part of the reading frame, which is capable of being translated. ORFs are defined as the continuous stretch of codons present between the start and stop codons. When the translation is initiated, the ribosome selects a reading frame starting from initiation codon and then translates the selected reading frame until it reaches the stop codon. Thus, it is essential to pay attention to the reading frames for proper translation to occur.
