To determine: The KM value of an enzyme with substrate concentration and reaction velocity.

Concept introduction: Enzymes are the biological protein catalysts that alter the speed of the reaction in a biological system. The chemical reactions that are catalyzed by these enzymes are studied under the enzyme kinetics. The reaction rate is measured based on the rate at which the substrate of the enzymes combine with the enzyme and form the products. KM is the Michaelis-Menton constant. KM value is not a direct measure of the affinity of the enzyme toward the substrate but the concentration of S, where the velocity at the maximum is half. Vmax is the maximum velocity with which the enzyme catalyzes the reaction. 

The rate of enzyme reaction (v0) =Vmax[S](KM+[S]).
