Question
You are studying SARS-CoVb. You hypothesize that upon entry to the host cell, the virus escapes from the endosome by inhibiting
endosome acidification. You are trying to determine which viral protein is responsible for inhibiting endosome acidification. You have recently read a
paper where they use pH-sensitive GFP to measure pH in different organelles. You decide to use pHlourin attached to an endosomal protein to
measure the pH in the endosome in the absence and presence of different viral proteins.
You first construct a standard curve using buffers and ionophores and get the following values:
pH
Emission ratio
5.5
6.
1.25
6.5
1.5
7
2
7.5
2.5
You then measure the pH in the endosomes and get the following data:
Sample
Emission ratio
pH
A
1.1
1.0
E
0.9
2.25
No protein
1.0

Expert Solution
Step 1

When a cell recognizes a viral genome in its cell body, endosomes are used to encapsulate and subsequently eject it. During this process, acid is released into the endosome in an attempt to destroy the viral genome, but this acidification can occasionally cause a shift in viral proteins, allowing the viral genome to escape from the endosome and into the cell's cytoplasm.
