The Bradford protein assay is a dye-binding assay based on the differential color change of a dye in response to various concentrations of protein. It is used to measure the total protein concentration in a sample. This assay makes use of a Bovine Serum Albumin (BSA) as a standard which is a globular protein. This is done by comparing an unknown quantity of protein to known amounts of the protein standard being used. The protein standard is measured, and its absorbance and concentration are plotted on a graph to create the standard curve. This curve is then used to determine the concentration of the unknown protein. The table below presents the standard calibration of the BSA standard for a study on freshly isolated brain microvessel of Sprague-Dawley male and female rats at a wavelength of 595 nm and pathlength of 1-cm at room 25°C.
