Principle
Specific gravity of the blood is largely determined by the amount of
hemoglobin it contains.
Materials
1. Copper sulfate reagent
2. Lancets
3. Cotton
4. Alcohol
Procedure
1. A single drop of patient's blood is placed in a solution of copper sulfate
with a specific gravity of 1.053 (Equivalent of density of blood with a
hemoglobin value of 12.5 g/dl)
2. Normal Test results occurs when drop of blood falls through the cop
sulfate solution without rising, indicating the blood has a >12.5 g/dl
hemoglobin level.
Precautions:
> Do not mouth pipet the Drabkin's reagent. It contains cyanide and is
poisonous.
Always use clean cuvettes.
Ensure that the dilution is made correctly.
Set the wavelength at 540nm.
> Mix the blood ten to twenty times before sampling.
> Do not squeeze the finger hard at the capillary puncture site.
